Canterbury scored 10 tries, including four to Milford Keresoma.
With Tom Taylor, Colin Slade and Tyler Bleyendaal unavailable, 19-year-old Richie Mo'unga made his debut at first five and scored 27 points.
The result moves Canterbury up to second in the Premiership, while Manawatu remain fifth in the Championship.
