The teenager finished with a three-under par 69 to finish 11-under, at the World Ladies Masters in Taiwan, to win $US150,000.
The 16-year-old had already won four professional tournaments before turning professional in October, but because of her amateur status at the time, she had to give up more than $1 million in prize money.
Ko finished three strokes ahead of Korean professional So Yeon Ryu with world No. 1 Inbee Park a further shot back in third place.
She will begin her 2014 campaign at the Bahamas Classic at the end of January before playing the New Zealand Women's Open in Christchurch.
