State Highway One near Balclutha was closed overnight after a fatal crash.
Police said a four-wheel drive vehicle and a car collided head-on in rainy conditions at Stony Creek, 10km north of Balclutha.
A woman died at the scene and another woman is in hospital.
Police expect to reopen the road later in the day. Diversions are in place.
