State Highway 6 will be closed until about 3.30pm on Wednesday following a fatal crash between Murchison and Nelson.
Emergency services are at the scene at Motupiko and diversions are in place via St Arnaud.
One person died following a collision between a car and a motorcycle.
Motorists are advised to avoid the route if at all possible.
